Brix measures the grams of dissolved sugar per 100 milliliters of liquid, and a refractometer reads it directly from a drop of kombucha. Converting that reading into the sugar remaining in a serving after fermentation takes two more numbers: a base tea reading and a serving size.

The base tea Brix reading, taken from brewed tea before any sugar is added, accounts for dissolved solids in the tea that are not sugar; it defaults to 0 when no base tea measurement has been taken. The raw Brix reading comes from the finished kombucha. Serving size, in fluid ounces or milliliters, sets how much of the batch the sugar and calorie figures apply to; common serving sizes are 8, 12, and 16 ounces. Calibrating the refractometer with distilled water beforehand, and keeping the distilled water, base tea, and kombucha samples at the same temperature, keeps all three readings comparable.

Adjusted Brix equals the raw Brix reading minus the base tea Brix reading, which isolates the sugar contribution from the other dissolved solids in the tea. Sugar in grams equals Adjusted Brix x (Serving Size in mL / 100). One fluid ounce equals 29.5735 milliliters. One teaspoon of granulated sugar weighs approximately 4.2 grams, and each gram of sugar provides 4 calories, which converts the gram figure into teaspoons and calories.

A finished kombucha reads 6.0 Brix, and its base tea read 2.0 Brix, giving an adjusted Brix of 4.0. An 8-ounce serving is 8 x 29.5735 = 236.59 milliliters. Sugar is 4.0 x (236.59 / 100), about 9.46 grams, which is roughly 9.46 / 4.2, or 2.25 teaspoons, and 9.46 x 4, about 38 calories from sugar.
